vanadium chloro peroxidase

project started in 1991

Vanadium chloroperoxidase (VCPO) from the fungus Curvularia inaequalis has also been a topic of research in the group for a long time. This enzyme has been characterized in detail using a variety of biophysical techniques. Its crystal structure has been determined and the enzyme has been successfully expressed both in Saccharomyces cerevisia (yeast) and in E. coli. VCPO catalyzes the two-electron oxidation of both bromide and chloride to hypobromous acid (HOBr) and hypochlorous acid (HOCl), respectively.
Our first directed evolution work successfully resulted in an enzyme mutant with a much higher brominating activity at slightly alkaline pH (pH 8).
